For a 3 panel brochure on 8.5x11, landscape, where should the 2 folds be?

That would depend on how you want your margins laid out and if you want to have an equal margin around each panel. In Word, you really don't have to worry about the actual numbers - it can set you up with a 3 column document pretty easily. First, go to File-Page Setup and change your orientation to landscape. Then, go to Format-Columns and tweak your settings there.

There are a lot of settings that you can tweak - you may want to leave the Equal column width box checked so you have equal panels, and if you need a visual as to where to fold, you can check the Line between option.

As far as the spacing thing goes, make the Spacing setting double whatever your left and right margins are, so that each panel will have equal margins all around. Of course, that's up to you, but it looks nicer if each panel was framed evenly all around (or at least it looks good if the left and right margins are the same, and the top and bottom are the same - top/bottom and left/right don't necessarily have to be the same).

Now, if you're looking for the actual measurements (that was pretty presumptuous for me to assume that you were using Word), you'd have to fold at 3.67" from the end.
