A small question that involves many answers at once.
There are many things that are considered the most important ones in Business:
First is consider which business you will be involved, doing a business plan that evaluates if this business is viable.
Then working into all areas of business ( Operations, Administration, Marketing, Sales, etc. ). Researching heavily on your core business and the products/services you'll be selling.
The customers are important, because you could have the best company in the world, but without customers to whom you sell your p/s there is no business at all.
Also, your competition must be in your thoughts all of the time. What you do and don't do will define if you lose customers or win them from your competition. Your competition will do the same from you.

The most important thing to know in business isn't really something you know. It's your drive; your determination; your will to succeed. We've all heard that If your "why" is big enough, the "how" will come naturally. It's true, and I'd say that it's the most important factor of success in general. But aside from that, I'd say that people skills are the most important. More specifically, knowing how people think, what motivates them, and how to persuade them.

#1 Resource: Local chamber of commerce (From there, customers, financing, moral support, legal assistance, etc.)
#1 Book: Atlas Shrugged (believe it or not)
#1 Key Indicator of Short-term Success: Cash (vs. revenue, profitability, balance sheet strength, etc.) (What you want to watch)
