first of all, mona lisa is a female. she's smiling because it was a portrait. don't you smile when someone takes a photo of you? leonardo da vinci was commissioned to paint a portrait of the wife of wealthy merchant francesco del giocondo's wife, lisa gherardini. this is the woman who is depicted in the painting.
i can't think of a better answer than that. perhaps leonardo was making a funny face.
