The "does" is contained in the contraction "What's".

In reality this sentence reads "What does he do?"

It's either, "What does he do".
Or "What is he do".
And considering I've said this before, it's "What does he do".
If you think "What do he do" is the way it should be said, you need a tutor, immediately.
