Officially, nothing! It's called a McGuffin, a plot device.

Unofficially, It's the diamonds from Reservior Dogs. Co-writer Roger Avery stated that the briefcase originally had diamonds. Tarantino loves to combine things from his movies to create a world, for example Red Apple Cigarettes.

IT'S NOT HIS SOUL! This is a very old rumor. He wore a band-aid because he had an ugly scar. Also, it's very funny how no one can come up with a biblical passage to back up the "Devil takes your soul through the back of your neck"

Tarantinos Answer

Tarantino had said that the first time he wrote a draft of the script the briefcase was going to contain diamonds (some say the diamonds from reservoir dogs) then he decide it to leave it open for interpretation... so the soul story is just an urban legend, although it is open interpretation, so if you believe in the soul story that's up to you....

Another thing, the actor that played Masellus, really need it the band aid ( i don't remember how he got injured) so its not like it was part of his costume

The case was originally supposed to contain diamonds, but they scraped that and just put in an orange light bulb. Tarantino has said that there wasn't an explanation for the contents. It's just used as a plot device to move the story along. I seen an interview with Tarantino once where he said that the idea that Wallace's soul being in the case was interesting, but he just wasn't creative enough to think of something like that. Still people speculate that it is still what's in there. Personally I don't care it's still a great flick.

Speculation abounds as to the nature of the mysterious glowing contents of the case:
Could it be Elvis's gold suit, seen worn by Val Kilmer (as Elvis) in True Romance (1993)?
The most persistent theory (most usually attributed to a friend of a friend who saw it posted on a message board by someone whose brother had read a report of a radio interview with Tarantino himself) is that it is Marcellus Wallace's soul. The story goes that when the Devil takes a person's soul, it is removed through the back of the head (this isn't part of any known religion, but this is what the message board posters say). When we see the back of Marcellus's head he has a Band-Aid covering the precise spot indicated by tradition for soul removal. Perhaps Marcellus sold his soul to the devil which would also explain why the combination to open the briefcase is 666.
Quentin Tarantino has said that the band-aid on the back of Marsellus Wallace's neck had nothing to do with an allusion to the Devil stealing Marsellus's soul... but that the actor Ving Rhames had a scar on the back of his neck he wanted to cover up.
Or could it be simply a 20-watt light bulb?
According to Roger Avary, who co-wrote the script with Quentin Tarantino, the original plan was to have the briefcase contain diamonds. This seemed neither exciting nor original, so Avary and Tarantino decided to have the briefcase's contents never appear on screen; this way each filmgoer could mentally "fill in the blank" with whatever struck his or her imagination as best fitting the description "so beautiful". The orange light bulb (projecting shimmering light onto the actors' faces) was a last-minute decision and added a completely unintended fantastic element.
In a radio interview with 'Howard Stern' in late 2003, Quentin Tarantino was asked by a caller the contents of the briefcase, and he answered, "It's whatever the viewer wants it to be."

I have heard several speculative explanations, as to what is actually in the briefcase, but there is one which seems to stand out in my mind, as being the more interesting poignant.

You recall the band-aid on the back of Marsailles Wallace's neck??? Some vague, rumored, albeit undocumented, unconfirmed and unsubstantiated biblical references reportedly dictate that when the devil takes your soul, he removes it from the back of your head. Now, if you recall, the combination of the briefcase is '666', and when anyone opens up the case, a brilliant, overwhelming light cascaded from it.

Is it possible that inside the case was Wallace's soul, and perhaps he was trying to buy it back??? The three kids in the apartment, who had the case (including ol' 'Flock 'o' Seagulls') were the devil's couriers; when one of the kids flew out of the bathroom, and fire a 'hand cannon' at Jules and Vincent, all of the bullets 'missed' them. "God came down and stopped these M/F bullets", because they were here to save Wallace's soul, hence the 'devine intervention' cognigtation.

An interesting speculation, to say the least, but as I said before, most poignant, would you say???

Wallace's soul. It was taken out of the back of his neck. That is why he had a bandaid on his neck.
