1. Capetown, CA to Eastport, ME = 3,580 mi (W- E)
2. Madawaska, ME to San Diego, CA = 3,249 miles (NE- SW)
3. Blaine, WA to Jacksonville, FL = 3,145 miles (NW-SE)
No highways are perfectly straight, but these three routes show the extreme distances from the SW, W, NW, NE, E, and SE.

The highways were deliberately designed to not go in straight lines so that drivers don't fall asleep at the wheel. And other places, they had to go around terrain features, or property of people who refused to sell, or whatever.
Although, for an answer, go with Bonneville Salt Flats in Utah. That is where they do the land speed records, and have had cars go over 600 mph in straight lines for miles.

The farthest I know of that is as straight as possible would be to travel I-40 from one coast to the other. It starts in Wilmington, NC and ends somewhere in southern California. I'm sorry, I don't remember the name of the town in CA and can't find the mileage, but I have actually driven it. Used to be known as Route 66, though parts of the actual route were changed slightly when it was converted to an interstate freeway.

The furthest trip possible would be from the north west corner of Maine to the southwest corner of California... i can't tell you right off the top of my head. The farthest north is Maine, farthest south i believe is Florida, farthest west is california and the farthest east is north maine also

Since the surface of the Earth is curved, you can't drive in a straight line across any significant portion of the US without either starting or ending at a very high altitude, or burrowing deep underground.
