I assume you mean the Art Institute of Chicago. Among Americans, American Gothic by Grant Wood is the most famous painting in the museum. Even if you don't know the name, you know the image:

It is a big place so the list is long, but they have a huge collection of impressionistic paintings, sargent, a pollack, lots of paintings from the rennaissance. I would visit their web site it could give you more info. It is a nice museum with lots of good art not only painting. I visit it often.
