It can be a positive thing. Like for instance if other people keep telling you that it's silly, or impossible to do something that you really want to do - but you decide to go ahead and do it anyway just to prove them all wrong. That can be fun. :-)

It means that the person has an uphill battle, a big struggle to get done whatever it is they are referring to. They may fighting for a cause that the majority of people are against. I'd say the comment is negative.

it means everyone else around her is against her/him/it. she/he/it is going against what everyone believes. sort of rebellion.
i see it as a positive thing. like that person is standing by what they believe in.
