Why wold you want to do that?
But, to answer your question, it wouldn't be hard, it's just like escaping from a car when you've driven into the lake. This is my worst fear, so I've taken classes. But, you can't pierce a hole on there, it would shatter, any glass would.
People want to pound pound pound on it with their fist, but it does no stress on the window.
You get an ice pick and put high pressure on the glass to make it shatter.

If you mean to drill an hole for an overflow you will have to empty the tank and purchase an expensive drill bit. If your tank (like most 55 gallon tanks) is made of tempered glass,it will shatter if you try. If your tank has very thick sides,it is probably "soft" glass and therefore drillable.

Common sense tells me this is a very, very bad idea. It somehow tells me to drain the water first before doing anything with the tank.
But if you must insist of being stubborn, please break out your tripod and Camcorder and record what you are about to do. I want to see some NEW bloopers on America's Funniest Videos.
