Sounds like you're doing tissue culture work. Water is hypertonic and would cause your cells to burst. Not good when you're trying to do an experiment!

Growth medium may contain inhibitors or binding proteins (if you have added serum) which could interfere with the amount of your compound that the cells are actually exposed to.

Ringers is an isontonic solution that relatively inert.

If you're talking about preparing dilutions for injection, Ringer's is an isotonic saline solution. Being isotonic, you don't need to add anything other than the Ringers to have an isotonic injectable solution.
