Biggest how? By volume, length, area?
Anyway, the Amazon has the highest volume, and the Nile is usually considered to be the longest, but the Amazon may actually be the longest... that's under debate.

While the Mississippi is the longest river in North America, it could have been longer.

If the river was what is now the Missouri-Mississippi, it would be the world's longest.
