Sprints, sprints, and more sprints. If you want to play in the secondary you need to work those fast twitch muscles and get them used to repeated uses in short bursts over a long period of time. Other than that, just work on your weight training overall and get stronger. It never hurt to be able to lay a hard hit on somebody if you're playing D.

Footwork, specifically the ability to not trip over your own feet when someone coming at you jukes one way or the other. Also, speed for keeping up with and running down offensive players, and upper body strength for making good open-field tackles.

Also, work on catching the ball. Knocking a pass down is good. But, an interception is better. You also may be called on to receive kicks or punts. Most borderline offensive and defensive starters start out on special teams and work their way up to their true position.

Speed. A last minute QB decision can wreak havoc on a DB. You have to see the field. In addition to working on your running and strength training, a good practice is playing "Monkey in the middle" and always volunteer to be the monkey.

In terms of football skills, learn to read the offense so that you aren't fooled by trick plays or caught out of position, learn to tackle(well) and work on speed drills.
