Typically delay through a device, i.e. latency is measured with a device known as a smartbits, made by a company called Spirent.

The smart bits is both the transmitter and receiver of the packet and the delays of the cables are accounted for.

If you want to measure latency over an entire network you need a device that can send a packet into the network that contains some sort of synchronized time stamp on it, which a receiving device can use to calculate latency through the network.
