A rubber-coated layer of fabric containing cords that run parallel to each other; extends from bead to bead and goes between the innerliner and belts of tread.

You have the tread on the tire, then steel belting which looks like close knit screen wire and directly underneath, you have layers of reinforced rubber which is called plies. Most lighter use tires are two ply while heavier pickup tires may be four ply and industrial tires can easily be 16 ply.
Plies, simply put are layers of reinforced rubber.

Ply generally refers to layers so it's probably the number of layers of rubber/materials used to make the tire. I would imagine a tire with a higher ply rating would be more durable.
