Most definitely Nightfall. I would probably suggest getting the Prophecies game of the year edition as well...I have all 4 campaigns (Prophecies, Factions, Nightfall and Eye of the North) and Nightfall is by far my favorite....The ability to create heroes in Nightfall also makes it a better game for those just getting into Guild Wars.

Factions is for PvP.
Nightfall is for PvE.
That said, I prefer Nightfall. I really like heroes a great deal, they make an irregular player with me have more fun.
Don't forget the original Guild Wars either. It has the longest campign of the three, but no unique classes.

I would say both. Factions is the easiest to grind a toon up to lvl 20 in, but with NIghtfall you get heros. If you have both campaigns, you can travel to NIghfall and get the heros with your Factions toon. If you include Prophesies, then there is yet another hero that you can get from there as well.
