Mine wasn't working and I was so sad...
I tried every possible way to get it working - I think I was at it for like an hour... o_0

Initially I get frustrated and upset. Then I find that I can do other useful and productive things without the darn computer using up so much of my time.

Short term= okay. Long term I'd probably go to the library to use the computers there.

I spend like an hour complaining and tyring to fix it while complaining since I"m the computer wiz in the family. Then I trash the project with the internet and just go on Microsoft word and start to write stuff ro I unplug my ipod and go into my room to draw or write in a notebook.

In 1983, Kary Mullis invented PCR while working at the Cetus Biotechnology, CA. Which eventually got him Nobel prize. PCR is a technique used in molecular biology, ofcourse PCR can be used in various fields including forensic (crime scene)investigations. PCR: POLYMERASE CHAIN REACTION You need to have some basic understanding about DNA and its structure before you go in to the details about the PCR. DNA is made of double strand of nucleotide sequnces, it is like a coil called helix (very simplified description). a small fragment of DNA can be replicated millions of times using PCR technique. This is a DNA amplification technique, where cloning is not neccessary, in cloning one must express and clone the DNA in a bacteria, but PCR eliminates those steps. The way the technique work is taking a single DNA fragment, one can rapidly make copies of it as many times as needed in a chain of reaction mediated by polymerase enzyme. How does it work: 1.First identify the DNA fragment of your interest, 2.Denature the DNA, by denaturing, the DNA double strand will open up and becomes single strands, typical temperature is at 94 degrees celcius (for about 1 min) 3.Add primers at 54 degrees celcius, primers will be moving around in the mix due to brownian movments, inonic interaction occurs between single strand primer and template (single strand of the DNA), the polymerase will start attaching and copying of templates. 4. At 72 degree celcius, this is the optimal temperature for polymerase, the nucleotide bases are coupled to the primer on the 3' prime end, and polymerase adds dNTP bases from 5' to 3' end. 5.A new DNA stran will be synthesized behind each primer. 6.Again DNA is denatured, primers are attached the DNA is copied double the amount now..., So, continue the reaction cycle (this is why it is called chain reaction), keep on doubling the number of copies of the original one DNA fragment in every cycle. 7. All the DNA molecules will have the region of interest. 8. one can run about 30-40 cycle, the automated pcr machine will take care of the cycles and temperatures that are critical. There are several uses of PCR: Gene sequencing helps to identify any defective genes, therefore, 1. In gene sequencing 2.Forensic DNA identification 3.Sex determination 4.Disease diagnosis 5.identify mutant genes 6.Gene therapy 7.drug targeting to genes, so, drug therapy 8.may be cancer detection (in case of gene involvment in cancer) and improving crop yeields, GM Plants 9. improving agricultural products in all these PCR can be applied. There can be more uses than the above mundane list.. A small amount of DNA from a drop of blood hair or any other body parts can be used to amplify the DNA into million copies by PCR.

I get upset and think of a way to get the internet working (I'm too addicted with the internet >_<). If that doesn't work then I'll do something that will forget about it.

I tried the library, but they kicked me off in 30 minutes. I went to a place where you can pay to use the computers and was very happy. That is how I coped. lol I got to do Yahoo, IM and emails. I was happy when I went home. : )

if mine's not working i usually try to do something else, or i could go crazy, lol. i'd watch dvd, tv, cook something, eat, cleaning up my room or the house, just anything gets me busy :p

yes i was getting yest error 999 pi@@ed me off and i had to wait 24 hours and it fixed self funny thing was it was only Y!A in the answering that i couldnt access & I was pi@@ed cuz i was sooo close to level 6 so i went over & did votes to get me to level 6 but it was long haul cuz its only 1 stinking point but i got there & back on today but i was reading when i couldnt get on
