It means that they think you are a little wound up or high strung in a way that it similar to the way someone on drugs would act.

speed is a type of drug that pretty much just gives you energy. But it is prescription so people who use without one act crazy. So, to make a long story short, when someone says, "are you on speed?", they mean, "are you on drugs?"

When they say that it doesn't mean that you are literally taking a drug, it just means that you act like it.
Not a very good thing to say to someone actually.

SPEED IS A SLANG TERM FOR UPPERS , DRUGS LIKE THE DIET PILLS OF THE 70'S & 80'S === WHITE CROSSES , YELLOW JACKETS , MOLLIES , BLACK MOLLIES , AND CRANK WHICH IS CYRSTEL METH. ( NOT CRACK BUT CRANK ) . ON A GOOD DOSE YOU COULD WORK A 16 HOUR SHIFT , THEN GO OUT AND PARTY FOR A FEW HOURS AND A FEW DRINKS TO COME DOWN OFF THEM .

I would think it was a teasing way of noting that 'you are so much more energetic than I am' or saying 'please slow down!' or maybe they have some reason to wonder whether you are really using amphetamines.
