yup = yes, yeah ,ya, ahuh, i understand, OR you can do something.

ex: Did you watch the game last night? Yup
ex: Do you like the color pink? Yup
ex: Michelle is such a brat. Yup she is.

Nope = no, nah, never, no way, your not allowed to, OR i do not understand

Ex: Do you understand the math homework? Nope
Ex: Do you know the answer to 75? Nope
Ex: Can you believe it?! Nope

Yup (yes) and nope (no) are American slang. The words are generally used when people are lazy in their use of language.

"Wanna go to the movies?"
"Yup."

"Nope, I'm not gonna go."
