Mel gave you a lot of good points, but to illustrate it can actually change a tragedy into a comedy by changing interpretation and infliction on lines, movement, facial expressions, etc. Whatever he tells the actors to do, how they dress, how the sets are done. Take for instance the fact that Bugs Bunny has performed lines of Shakespeare. The director animated it, took liberties with the material and made it funny. It's all in the delivery. Bugs obviously didn't do this of his own free will, he's a cartoon. He had direction.

The director has a huge impact on a piece of work. You can have them same story made in 50 different ways from 50 different directors, simply by the way they interpret it.
It is directors choice to choose things like different camera angles, lighting/sounds (digetic and non-digetic), costumes/props, and things like adding additional scenes. The way they interpret and create such elements can get messages across, and help us, (as the audience) to get a better understanding of the characters (their emotions, their status, or their relationships between each other) or of what is going on.
