As the computer is booting press your "F8 Key" repeatly which should bring up the "Windows Advanced Options Menu" . Use your arrow keys to move to "Safe Mode" and press your Enter key.
After into safe mode,run an anti-spyware to remove the spyware and adware on you computer.

If the alert is coming from your windows security suite then Yes you would remove it.
If the alert is coming from the web then you've got a problem.
Your pic was a bit hard to see...
If you are using a proper security suite that includes antivirus, antispyware, firewall, etc.. and is from a reliable vendor (like a mcafee, norton, etc) and it is properly configured and completely up to date.. then to ignore a warning is unwise.
If you are not sure... alt+f4 out of IE and run a complete scan from the security suite.

That window that pops up is what you call a virus or trogan, it infiltrates your computer and tells you its infected so that you buy some fake software and they steal your personal information. Click no every time and go find a real spyware remover and a good anti virus. I use ad-aware and avast anti virus personally. Both are free too.

Go to http://free.grisoft.com
and download the anti-spyware free edition. Then run a full scan to delete those spyware.
