Maybe you could tell him anonymously through a letter/note/email and he could figure it out without you ever "really" saying anything. Then you can lend him a shoulder when he needs you most.

Well, he WAS your friend before you told him. I guess that is my way of saying, yes, you should tell him, alone, away from people, because he will take it hard or deny it. Chances are the friendship and rooming together will end, so look for another place. I sympathize with your plight, and with him.
Alternative, and better, convince his wife to tell him and tell her you will tell him if she doesn't. That is what I would do. Offer to be around when she tells him, that way you will know it will get done and you can help with the fall out.

If he doesn't work too far away, when you see her enter the neighbor's, you could call him at work and tell him to come right home, it's an emergency. Then he could see for himself.
Otherwise, yes, tell him. I would. If he's as good a friend as you say he is, he will believe you. Just say to him, you know it's not your business but he's a good friend and a good man and you thought he had a right to know. Imagine if she brought home a disease or something or became pregnant? You would feel guilty.

Assume that he will hate you (or at least resent you and feel akward around you) and never talk to you again. Then ask yourself if you should tell him? Try to prepare a net for him. Will he need to stay with you or somebody else? Can you check on him to make sure he keeps going to work? You Break the news you buy the fallout.
He will hate you more if he finds out you knew and never told him.

Yes, you should really tell him. Especially since he is a good person. It is only going to make matters worse if/when he finds out he was being cheated on. If he finds out you knew he was being cheated on and didn't tell him, he'd probably be upset.
*Or...you could "accidentally" walk in on them, so your friend will find out for himself. Haha, just kidding..

Find a way to discretely ask him if he wants you to tell him. Ask maybe, "Is ignorance bliss?"
Theres a song by Mario, "I don't want to know" in which he talks about how he doesn't want to know his girlfriend is cheating on him 'cuase it'll hurt too much.

I would tell him. Telling him may hurt his feelings but not telling him will probably cause him more pain in the long run.
