Depends on the users actually.

Most users don't notice it at all, until it breaks their system and/or annoys them to a point.

The most advanced users immediately notice the loaded programs on startup that they don't recognize, notice the RAM usage getting high. As well as the frequent disk access that happens.

Asking a question about how long it takes to realize your computer has a virus, you are looking to destroy your pc.

You should run your antivirus/antispyware every time you use your computer. Ofcourse after you have completed use, before you shut down.

You want to destroy any virus, or viruses, before serious damage is done to your system, or your data is stolen by a hijacker.

If its a trojan or rootkit you may never notice it, others simply corrupt your system on contact.
