Dallas Cowboys vs Washington Redskins, it's lifetime rivalry. When the league rearranged the divisions everyone was agreed to not touch the Dal v Was rivalry that the two teams had to be in the same conference. When these two teams meet throw out the current records anyone can win. It's the ONLY game left on Dallas's schedule I'm worried about this year!
Anyone saying Colts-Pats has a very short term memory.

New England Patriots vs Indianapolis Colts
Why? Because they have the one characteristic that makes a good rivalry. They're both at the same level. If you have a match-up like the Patriots vs Dolphins, how can you call that a rivalry? But the Patriots and Colts are evenly matched up as the two best teams in the NFL. When they match up against each other, the victory could go either way. It'll be a close victory until the end, which will eventually depend on clutch accuracy. Tom Brady and the New England crew has beaten the Colts by a close win on numerous occasions and vice versa on Peyton Manning and the Indianapolis crew's part. It's exciting to watch a Patriots vs Colts game because of the suspense in who will win. You never will know, that's what makes a good rivalry!

Well, everyone says its Pats-Colts but there's no historical basis, it's media-built. They delibrately schedule those teams because of their talent and records.
I prefer geographical or division rivalries, because no matter what the circumstances (talent, in-coming record, media, etc) the game is important. Bears-Packers is important. All of the NFC East games are important. I've never seen a division with more passionate fans (Giants-Philly, 'Boys-Skins, Giants-'Boys are the best).
Colt-Pats has the all the shiny fixings now, but 5 or 10 years from now when either one of these teams will suck, it won't have any basis in the NFL. The ones I mentioned earlier will always be significant.

one of the best in the past has been Cowboys-49ers, but seeing as the boys would whip up on them every year of late, so right now the pats-colts seems to be a good rivalry. and it has got better since a couple years ago

I don't think the NFL has a real rivalry. They have media hyped rivalries, like Brady vs Manning or Belichick vs Mangini, but no real rivalry like in hockey with the NY Rangers vs NY Islanders or Calgary Flames and Edmonton Oilers or any world soccer derby where the fans want to kill each other and they designate areas for away fans as well as players hating each other.

Because it's one of the oldest rivalries and between two very passionate cities--I'd have to say the Giants and Eagles.
I also like the recently revamped Browns and Steelers.
