anything smaller than 500-600 and you'll be bored in a week, not to mention ninja 250s are about the gayest looking thing in the world!
take a class (it's worth it) if you don't already know how to ride, they'll put you on 250s and that's about all the time on those you'll need.
I would hope you know this: say to yourself "this is my first bike, odds are that at the very least I will drop the thing at least once in the next year so I don't need something expensive"
Personally I started on a 1990 vfr 750, the bike looked better than my 03 gsx-r 1000 if you ask me (I had the vfr last year). It's made by honda so it lasts a long time and if you get an older one it won't have enough power to kill you right away, just right if you ask me, under 100hp. less than the new 600s. The reason I would recommend the vfr over the cbr/r6/ninja other than the looks is the insurence. The VFR is classified as a cruiser (although it looks like a sportbike, just look at the newer ones) making the insurence insanely cheap (under $100/yr. for a 20y/o with a (near) perfect driving record.
not to mention I got the bike in great and I mean exceptional condition for under 3k. and it was very comftrouble. (sp)
I would also aguree with the guys that recommended a suzuki sv650, but those are naked (no ferrings like a harley), I don't know if your going for the street look or not?, just make sure it's older, if it makes it through the first year an old bike won't depreicate much more and you can always upgrade.
That's my two cents.

Kawasaki EX250 or EX500, Suzuki GS500. All will get 50+ mpg, are dependable, well built, and if you get a used one you can probably ride it for a year and then sell it for what you paid for it when you're ready to move up.
More importantly, you're a beginner. You want something that is forgiving. Low weight and limited power are your friends at this point. For the same reasons the military doesn't train their pilots in F-15s, you don't want a large, powerful bike.
If someone tells you you can start out on anything over 500ccs and that it's just a matter of just being careful with the throttle, ignore them. You want to be confident and relaxed, which is difficult on a bike that can instantaneously put 60 to 100 horsepower to the rear wheel. That rules out an SV650, or any of the 600cc sportbikes.

If you have never ridden dirt bikes or motorcycles before I would highly recommend the kawasaki 250. It's cheap and real easy to learn on. The only problem is that you will most likely want more power within a few weeks or months (however fast you learn). So whatever you do, DO NOT BUY A BRAND NEW BIKE for your first bike. Btw, I'm not sure, but someone told me that those 250's get like 70+ mpg.

2008 Ninja 250 if your ego isn't bruised because of the smaller displacement than a typical squid "requires". Accelerating fast in a straight line is easy on any motorcycle, no skill in that anyway unless you're doing it competitively at a drag strip. Economical in all ways at under $4K out the door and 50 mpg. If you're average U.S. male in height though the older generation was a bit cramped, not sure about the 2008.

The most important thing you should do before purchasing any motorcycle is to look for the very best motorcycle training school in your area. It doesn't matter whether it's a Vespa or a full tilt Harley, you will still have the same opportunity to be dead or maimed unless you know how to get yourself out of the simplest but most often fatal of situations. I know, I spent eight days in hospital.

I've always recommended Ninja 250's for beginners. About 250CC and they cost like 3,500 bucks so if you wreck then hey at least you didn't wreck a 10,000 dollar bike.

Look at Yamaha Virago 250. It looks good and makes good power.
