Yes it does run just fine in Vista, however you will need to make sure you're running it from the Administrator account or enable one on your system for the patches to install and work right.
I have not personally had this problem, but others that have Vista on their systems have said they had trouble with it.
Also, Vista is a HUGE memory hog, so hope you have a lot of memory in your system so that you can run it ok

Yes, although since Vista takes up a lot of RAM, you won't be able to get by with minimum system requirements.
