When the sea turtle becomes an adult, the only way to tell if its male of female is by the length of the tail. Males have a long thick tail while females have a very short tail.

Hatchlings are tough. Your best bet is to look at the position of the vent. If it is under the carapace, you probably have a female. If it is beyond the carapace, a male. The difference gets more pronounced as the turtle matures.

The best way to sex a turtle is to put down some food. If he eats it, you have a male. If she eats it, you have a female.
