'And' is a conjunction and doesn't need to be pluralized. (I'm not sure I understand your question)

For your other question, I would say: How many times is the word 'and' used in the sentence?

It is categorized as a "coordinating conjunction" and in fact all the coordinating conjunctions (FANBOYS) can't be pluralize.
