Your question has attracted some rubbish answers. Let me explain.
All railway networks need to have their tracks checked on a regular basis. (In the days of British Rail, every peice of rail was checked every third day.)
For safety reasons, these checks cannot be done during periods of intense operation. Moreover, checks in tunnels demand greater safety requirements.
Even then, when faults are discovered, time must be found, within the constraints of an operational timetable, to undertake the neccessary repairs.
For these reasons, it is necessary to close the London Underground each night. Last trains leave central London around midnight, with the last trains stabled in their depots by 1am. The first trains of the morning are staff trains, leaving depots around 4am.
It is possible to plan for a 24 hour service in advance for special occasions. For example, in recent years, there has been an all-night service at New Year.
If the Underground system was 24/7, then it is highly likely that within a month there would be a serious tunnel derailment.

The last trains leave their first stop between midnight and 12.30am. They will be scheduled to reach their final station by 1am.
It's usually taken the there will be no passenger trains running between 1am and 5am.
In practice, last trains of the night reach their final station just before 1am. The first trains of the morning leave their first staion just after 5am.
