Both of my hands are all of the sudden locking up. I haven't done any strenuous activity other than walk around greeting people at my job tonight. What could this mean?

If by greeting people, you mean shaking hands - it's possible that you have some kind of muscle fatigue, the sort that occurs whenever you give muscles a workout they are not used to. In that case, you may also be experiencing some pain or tenderness in your forearms, which is where most of the muscles involved in the "grip" of your hand are located.
I'd say what you're describing is similar to the well-known "writer's cramp". It should go away with rest (warmth and NSAIDs like aspirin or ibuprofen will help the immediate discomfort).
