You know how chromosomes look like clothespins or butterflies? The two strings are the sister chromatids and they are held together at the centromere. Together, the two sister chromatids make up one chromosome.

One of the sister chromatids is an original strand of DNA. The other sister chromatid was formed by copying the original strand. This happens during replication in the S phase of interphase. So the two sister chromatids are identical. They have exactly the same alleles.
