This is a somewhat graphic colloquialism denoting that a person is "Uptight" ie nervously concerned and worried about something, to the point of being over-sensitized, and subject to some severe reactions if provoked on some subject. I believe it is American usage not British but am not sure.
Telling you this suggests he would greatly appreciate it if you (and your cohorts) would not broach the subject with him----ie "mind your own business", or "lemme be!" or "Hau ab"
These are guesses----the truth would become know if you would have said "I beg your pardon !?" showing slight indignation but not too much if he is a Professor.........RHIP !

In Michigan, USA, if someone said that it would be slang and simply mean that he was stressed or anxious about everything, unable to let even the little things go. Someone who is not at ease is said to be "tight-assed" or as your professor stated. Tell him to speak the Queen's English if the slang makes you uncomfortable.

Was he an arts/humanities professor, in particular, in the performing arts? If so, perhaps he was stoned.
Maybe he actually said to himself, "I have a tight sphincter, but I wouldn't wan't to admitter, or I'd let everyone know I have to get to a toilet quickly for a **itter."
