I do not believe he was. I believe he remained a bachelor. I am 99.9% positive he did not marry. (I gotta leave myself some wiggle room!)
I was reading something about the Lincoln assassination just recently, and it went into quite a bit of detail about Booth and his co-conspirators. While I had already known that Booth's older brothers and father were also actors, I didn't know that he was, by many, considered the best actor of the whole, talented lot.
I know he was considered extremely attractive, and that he was very popular, so I would imagine there was quite a bit of female attention thrown his way. My guess would be that if he had anything, it would have been a mistress, or maybe several girlfriends. He was extremely interested in politics and especially the whole issue of slavery from a fairly early age, and I think he probably put his energies into his political and stage life, rather than romance.
As I think about it, I realize I have read a lot of things about him, and there has never, ever been mention of a missus for him, so let me change that 99.9% to a 100%. The guy lived his life as a bachelor, and died one, as well.

John Wilkes Booth was married twice. Booth met his first wife at a dance and in a fit of mutual passion; they were married the next day. He married Izola Mills in January 1859. Nine months later, on October 23, 1859, a daughter was born. Her name was Ogarita Bellows. Izola was legally married to Charles Bellow when she met and fell in love with Booth. Because Izola was still married to Charles Bellows when she married Booth, Ogarita received Charles’ last name.
On April 25, 1865, Union soldiers in pursuit of Booth were closing in on him. Two of Mosby’s men rode up to Booth and warned him that soldiers were approaching. Booth left the Garrett home and escaped capture. Union soldiers killed a Confederate soldier in the Garrett barn and he was mistaken for Booth. Powerful men in Washington wanted the massive manhunt to end. They declared the Confederate soldier to be Booth. The manhunt was over and any involvement they had in Lincoln’s killing would remain undiscovered.
A year and a half after Lincoln’s assassination, Booth was living in San Francisco. Izola went to see him and became pregnant. Izola’s second child was born on February 22, 1870. Izola and John Wilkes Booth became separated and would never see each other again.
Booth wandered until he found himself in Sewanee, Tennessee. There he met and married Louisa J. Payne. A justice of the peace married them on February 24, 1872. About nine months later, Louisa gave birth to Laura Booth.
