It does not affect the retention time, but if you inject a big blob of something, the resolution will suffer.

Ah, a topic near and dear to my heart.....

Sample size does not affect retention time unless you overload the column. Then the peaks will be blown off scale large and tail off well after the main peak. The main peak will be a bit later than usual simply because there is too much sample for the column to accomodate.
