By default Vista is set to defrag automatically I think once a week. If you right click on your hard drive and the go to properties and into Defragment now... There should be an option there that tells you whether auto defrag is on, how often it is set to run and when the last time it ran was.
You should defrag once every week or two. It's probably easier just to let Vista automatically do it as I know I'd never remember otherwise

The built in defrag in Vista seems to be one step back than XP as it doesnt give any info on what it's doing. If you set it to defrag to a schedule it will check to see if defrag is necessary and work in the background if it is. If you do a defrag manually you have no idea how long it might take. Best to set it to once a week and forget about it.

You can schedule a defrag. for any time.....mines every week at the moment, but you can also defrag manually at any time, in fact, mines defragmenting right now whilst we speak. it doesn't slow the system down at all, it just works in the background.
