Well, since homo- is a Greek prefix (meaning "same"), not a full word, the word isn't very specific. It could mean homosexual, homo sapien, homogeneous, etc. "Homo" is most often used as a derogatory term for a homosexual person, either male or female. Many homosexual people find the word "homo" offensive.

"Gay" also has multiple meanings. The original meaning of "gay" was an adjective meaning cheery and high-spirited. However, "gay" is most often used in its noun form, as slang for a homosexual person, either male or female, but usually male. Homosexual people usually don't find the word "gay" offensive, but it depends on the tone of voice of the person who used the word.

Homo is a shortened version of homosexual, meaning one is attracted to the same sex.
Gay is a slang term that I presume came about because when you see groups of Gay people, they always seem happy, if not rowdy.
The first time I heard Gay used in that context in a pop forum, was when Cary Grant used it in an old B&W 1930's movie, though I'm sure it's older than that.

"Homo" is a negative term for a person who is homosexual. It's used as an insult. "Gay" is a neutral term used to refer to a homosexual person -- no insult is intended by its use. So the difference is not in literal meaning but in the emotional meaning of the words.

That's a good quetion.
It's the same thing in todays english, but back in the middle ages and colonial times, Gay was to be happy and bright, not to be someone who has sex with the same gender. Now it means the same as a Homosexual.
