Well, I live in Falmouth so I suppose there's a slight conflict of interest there :)
Woods Hole (within Falmouth) really is an amazing village, I highly recommend it. It is mainly comprised of three groups: affluent heirs, intellectual scientists, and artsy teenagers. Very quaint and free-spirited, mostly.
If you go to Falmouth, go to West or town central.
Sandwich has a bit to offer, not too much though.
Chatham is great... if you're willing to spend a nice buck. I'll give it to you that it is very pretty and a generally well-marrered place; however, be prepared to loosen the purse strings here. Fabulous hotels and restaurants if you can afford it, and gorgeous beaches.
If you want the whole Cape Cod experience, Brewster is a pretty good bet. It is relatively affordable and caters very much to the tourist population. I reccomend Ocean Edge Resort for a place to stay.
Of course you have to make the trip to Provincetown, on the tip of the Cape, for a fun day or two. It has an extremely gay population and many cool, different shops providing you with a laid-back atmosphere.
Truro has nice beaches but not much else... literally.
And Martha's Vineyard and Nantucket are awesome if you want a fairly expensive alternative too. Nantucket more so; I reccomend MV. Fairly same deal, they have excellent restaurants and quaint shopping, etc.
All in all, I would say go to Martha's Vineyard or Chatham if you are willing to spend, and Brewster or Falmouth for a slightly more reasonable alternative.
Watch out, the Cape's highly addictive!

It's funny, some people in Chatham have bumper stickers that say "Chatham: a quaint drinking village with a fishing problem"..LOL..so, yes, Chatham is one of the more quainter seaside villages down here...Osterville in the Mid Cape area is nice too, and Ptown is def on the list...

I really like Brewster. I'm partial to the north side of the Cape. I like the general store in the center on 6A. And I like the farmer's sign (put up for the benefit of tourists) letting people know that trespassing isn't advised because his bull can run 100m in less than 10 seconds. The lights on the houses in Brewster are beautiful this time of year.

I grew up on the Cape, and while Chatham is the obvious tourist answer, I think you'll do better in Brewster/ Welfleet/Truro/PTown if you have a car. And it will likely cost you less to stay in one of those towns.
