In 1700, Boston was the largest city in the English, American colonies, with a population of 7,000 (mostly) Englishmen. New York had a population of 5,000 (mostly) English and Dutch. However, New York had a farily large population of slaves which were, to the best of my knowledge, not included in this figure.
By 1720, Boston was still the largest city (12,000), followed by Philadelphia (10,000) and New York (7,000).
By 1790 (no longer the colonial period), the major cities were Philadelphia (42,000), New York (33,000), Boston (18,000), Charleston (16,000), and Baltimore (13,000).
Several other smaller cities (under 10,000 by 1780) were important colonial capitals and centers of trade; cities such as Providence, Albany, Newport, Savannah, Norfolk, and Hartford come to mind.

all the main appropriate cities (NYC, New Orleans, Charleston, Philadelphia, Boston) have been on the mouths of rivers ideal out to sea. This allowed them get admission to to commerce in the interior, and out to sea.
