I do not know what is the best way, I live quite far of, in the Netherlands, but I looked for you on the train planner for France.
If you leave Lourdes 11.14 on the Saturday you will arrive in Roma the next day at 9.42, long trip and expensive too, most likely.

I have seen that the part Nice to Rome takes as long or longer than traveling by train, so I assume that the whole travel will take more time. But likely less money.

The flights all seem to have a stop somewhere, or even two, and seem to take half a day, at rather high cost (937.65 € ) but I am sure that if you use the train to a near town and fly from there you will get a better price.

Toulouse to Rome has tickets from 100.03 € leaving at noon and arriving in Rome by 17.30, there are more, more expensive, flights, so you are not restricted to one line only.
