It completely depends on what you're in too. If it were me, and I was just turning 21, and wanted to experiance all that age has to offer... hands down I would pick Las Vegas. It will blow your mind. How long are you staying (I'm assuming between flights)? There are so many clubs, bars, casinos, women, etc all in one area it's amazing. Also, it is probably easier to fly into Vegas than San Fran (due to fog, wind, etc) and you would be less likely to have delays. Now, if you stop in Vegas and want to do ANYTHING, it will cost you, remember that.
If you're more into a relaxing stop, seeing beautiful sights and stuff like that, by all means stop in San Fran. It will be cheaper than Vegas, and there are also plenty of things to do there. It's a lot more relaxed then Vegas, and the people generally are very nice there.

In Vegas there are a few things to do (gambling, drinking, etc) and, yeah, its fun. But in San Francisco there is SO much to do. Trust me, especially in the summer, San Francisco is the better choice.
