Hire a professional who will use humane traps. Good for you to not to use a poison. Do not try this yourself. Squirrels can carry rabies and you do NOT want to risk getting bitten. You will need to also find how they got in and cover the holes they used to get in. The professional will know exactly what to look for in an entrance point and most likely will also be able to block any entry points. The most likely places they use to get inside are the under roof vents, usually covered with a screen, which they loosened or was already loose.

Well, you get both you hands, (the right and the left) and move them towards the squirrel. Once you see the squirrel, and have full control of at least two of your hands, advance them towards the fluffy brown creature and clinch your hands into a grasping position. Once this is done, open a window and hurriedly let do of the fur ball while swinging it towards the opened hole in the wall. Congratulations, you have just removed a squirrel from your attic!!

You can also trap them, but you would be best to get a professional to do it. They can tell where they are getting it, and put a cage with yummy tasting treats in side it that will lock once the squirrel is in. As for the poison, the stuff they use nowadays kills them fast and as humanely as something can be killsed and dries them up in days so they don't smell or decompose the regular way.

Be prepared to close up their entry, fix the exterior wood work, if you go into the attic personally to scare them out...
Or Place a trap cage, some exterminators will rent them to you.
Then let them go in a park...
But you still will need to close the exterior of the house.
Exterminators cannot place poison for them, it has to do with the type that is effective, being a sexual reproduction inhibitor...

When you get a good answer pleas let me know. You are not along. When we remodeled are home we found nest of them in the floor of are Attic above are little girls room. That scared the hell out of me. We thought we got them all out by using traps but they are back again this winter and cannot get to them. They are smart, and I'm scared that they will get down into the main house.
So when you find a good answer pleas let me know.

First you have to close up all the holes that let the squirrels get in, then set some havahart traps and put the squirrels outside. I tried using window screen to close holes but the squirrels cut right thru; I had to use hardware cloth to keep them out.

Get a have a heart trap. You never have to touch them. They may have babies & killing things is wrong, especially innocent things. But I definitely know how you feel.
They are pricey or you can get one from the local shelter to borrow. They get in the trap & then you pull the lever to let them go.
Do you have a local animal lover? Maybe a cat lady? They are a huge help. Or maybe a vet tech from a local vet clinic?

Get several boxes of mothballs. They are cheap at a dollar store. Scatter them around in the attic(wear gloves). They usually run off any rodents, they don't like the smell. I had a field mouse problem a couple years ago. Moth balls ran them off. No muss, no fuss.

My sister put traps in her attic. Each time she caught a squirrel she would take them to woods away from her home and release them.
