Luke Richardson was drafted seventh overall in the 1987 Entry Draft by the Toronto Maple Leafs. That year, he made his NHL debut and then scored his first goal on December 16 against the Red Wings. In 78 games that year, Luke scored four points. After being limited to 55 games the following season, Luke then scored 18 points in 67 games during the 1989-90 season. In 1990-91, Luke scored ten points and accumulated 238 penalty minutes in 67 games.

On September 19, 1991, Luke was traded with Vincent Damphousse, Peter Ing, Scott Thornton, future considerations and cash to the Edmonton Oilers for Grant Fuhr, Glenn Anderson and Craig Berube. In his first season with his new team, Luke scored a career-high 19 assists and 21 points. In the playoffs, he helped the team reach the Campbell Conference finals before losing to the Blackhawks. In 1992-93, Luke played 82 games and tallied 13 points. In 1993-94, he played 69 games for the Oilers.

In the shortened 1994-95 season, Luke scored 13 points in 46 games for the Oilers. The following season, he played all 82 games and won the Oilers' Community Service Award. In 1996-97, Luke played all 82 games and helped the Oilers make the playoffs for the first time in five years. In the playoffs, he helped the Oilers reach the second round.

On July 23, 1997, Luke signed as a free agent with the Philadelphia Flyers. That year, Luke played 81 games on the Flyers blueline. In 1998-99, Luke played 78 games and tallied 116 hits and 94 blocked shots. In 1999-2000, he improved to 125 hits and 109 blocked shots as the Flyers won the Atlantic Division title. In the playoffs, he helped the team reach the Eastern Conference final before losing to the Devils. In the second round against the Penguins on May 4, Luke assisted on Keith Primeau's overtime-winning goal in what was the third-longest post-season game in NHL history (132:01 minutes).

In 2000-01, Luke led all team defencemen with a +23 rating as the Flyers reached the 100-point mark for the second straight year. He was one of two Flyers to play in all 82 games and one of three defencemen on the team to score a short-handed goal. On February 1, Luke's teammates and team awarded him with a Harley Davidson for playing his 1,000th career game.

In 2001-02, Luke played 72 games for the Flyers and collected nine points, 113 hits, and 83 blocked shots. The Flyers, meanwhile, won the Atlantic Division title. In the playoffs, Luke played five games in an opening-round loss to the Senators.

On July 4, 2002 Luke signed as a free agent with the Columbus Blue Jackets. That season, he played all 82 games and scored 13 points. In 2003-04, Luke played 64 games on the blueline. The Blue Jackets, however, dropped seven points in the standings and were amongst the bottom-five teams in the league for the third-straight season.
