I'm insulating my home office with pink rolled fiberglass insulation. I've heard that insulation is more effective when it's not compressed, so should I fluff and spread it before I place it?

Dead air movement, that is the goal. When insulation is compressed it cannot trap air and the "R" rating is reduced.
Do this little experiment. Place uncompressed insulation next to a heat source and measure the temperature of the air on the reverse side. Now compress that insulation and test the temperature again. Thermal pane window is trapping dead air between two sheets of glass. You did not mention if the insulation was wall or ceiling, but in any case be sure to deal with vapor barriers and condensation.

I have to agree with one answer who states it should be Ok when unrolled. Certainly you can determine at the point of purchase it it seems compressed already; but once unrolled it will bulk out some. It sounds like it isn't paper sided.

When installing it between studs just take a wide blade paint knife and press in the edges. The drywall; or whatever choice of material you use; will possibly compress the batting to the thickness of the studs 3 & 1/2 to 3 & 5/8 anyway: IF the batting is an R value that dictates it's thicker than that when fully fluffed.

It should fluff enough when you roll it out.
The compression statement is when you are stuffing it in tight areas.
