My favorite of all time would be Mel Brooks' Blazing Saddles. I highly enjoyed the remake of 3:10 to Yuma, and Joss Whedon's Firefly series and Serenity movie.

My all time favourite is Rio Bravo, it has everything you want from a western. Close behind it would be The Searchers, a fantasic performance from Duke Wayne.

Serenity by Joss Wheedon of Buffy fame. It's set in space and is the best 'Western' ever written and given a contemporary twist.
