You need good anti-virus software, anti-spyware and a firewall. You can get all these together with an Internet security software suite. Your computer may come with one of these already installed. Keep the security software suite current by doing updates with the vendor. Most security software has an automatic update feature, I would schedule it to check for updates daily. I would also automate is to scan for virus' and spyware daily, at a time your not going to be using the computer - i.e. middle of the night. The computer has to be left on for this to work.
Get a program called hijackthis and run it and create a log first thing with your computer. If you ever do get malware or some other nastyware you can use this first hijackthis log to determine what has changed from when the computer was a virgin. You can download hijackthis for free, just search google for it.
Never give out your main email address on things like surveys or contests. Those are just scams to get your email address for spammers. If you want to do those things use another email address, such as your yahoo.com address.
Learn how to do system maintenance, such as cleaning up the history, cache, file system, doing a disk defragment.
Do regular backups of your personal data. Personal data is things like music files, pictures, documents etc.. Nothing sucks worse than having a hard drive fail and losing $100's worth of music files or pictures that can't be replaced. For that I'd recommend an external USB drive. You can get a 200-300Gb external drive for small money, it's worth the investment. If nothing else, back up to CD or DVD. Do this regularly. My wife is clueless on this, so I got some software to automate the backups, every Thursday her data is backed up while at work.
Like a car, if you take care of your computer it will serve you well for many years.

Well, after you purchase a new computer, it will usually come with a free trial of anti-virus on it.
Check the internet regular or ask the people around you to see which anti-virus is good for your computer.
To take care of a desktop, don't install too many softwares that you don't need, use the Disk cleanup once a week, and remember to clean the components inside of the desktop also.
