Check the BIOS and make sure the USB ports are set correctly. There could be a connection problem on the motherboard where the USB connectors are attached - just trace the wires to the motherboard and see if it connected ok. Power down first - of course!
You can purchase a PCI USB 2.0 card that will give you ports out the back of your PC.

Have you tried....
Control Panel > System Icon
Click on the Hardware tab
Click on the Device Manager button
Look down through the list of installed items for the USB section
Click the + to expanded the section so you can see all the devices.
Right click on and uninstall every device in the USB section.
Reboot the PC and it will reinstall the devices.
Often that will start a port working again.
