Blood cells are placed in a nutrient solution and treated with a substance that makes them start undergoing mitosis. They are then treated with a drug that blocks mitosis at metaphase.

After a period of time, the cells are fixed in a solution that makes them swell and become very fragile. The suspension of cells is dropped onto a microscope slide so that the cells break open, and the chromosomes stick to the slide.

The chromosomes are then stained and looked at and photographed. The chromosomes are cut out of a picture and arranged by size and banding pattern to give you a complete karyotype.
