Depends on what kind of shape you are in. 4 miles per hour is walking fast (15 min/mile) and unless you were in very good shape I doubt you could do that for the 12-14 hours a day you'll need to do the trip in two consecutive days.
Downtown to downtown as the crow flies is only about 80 miles, but as most cars go it's about 92 using I-94. Since you won't be flying, and you can't use the interstate, you'll have to use regular roads, so you'll do better than the interstate route but worse than the crow. The route I draw out below is 86.3 miles, according to Google Maps. At 4 mph, that's about 21.6 hours; at 3 mph it's about 28.75 hrs. So it can be done in two long or three shorter days. The simplest street route (you can't use the interstate)would be from downtown to use LaSalle, to Clark, to Ridge to Green Bay, to Sheridan in Highwood (green bay doesn't go through the northern part of Highland Park), back to Green Bay, into Wisconsin where Green Bay becomes WI-31, which merges with WI-32 in Racine and goes all the way into downtown Milwaukee.
BTW, this route will put you right into the middle of some very sketchy areas in Waukegan, and possibly in North Chicago too. I don't know what the route in Wisconsin would give you.
Are you planning a fund raising walk? Be sure to drive the route first so you know what you're getting into.

Its roughly 90 miles between the cities. So assuming you don't get ran over by a car I think it would take someone between 18-30 hours (you walk between 3-5 mph depending on your pace).
Just an FYI there is a train that goes from Milwaukee and its southern suburbs to Chicago and its fairly cheap if I remember.

Make it easy on yourself; just pencil in 3 days for the trip and you'll make it. All this nonsense using the r/d=t formula is just that nonsense . Unless you have somebody with you that will keep you walking at a very strict pace your actual distance walked per hour will vary on an hour by hour basis

Say it's about 80 miles and you can walk 3 miles per hour.
If you are uninterrupted by terrain, weather, need for sleep, etc, you could make it in about 27 hours.
Personally, I would take the train. It's faster and safer.
