IMO, the Canadian side is better, more to do. Since you're staying on the American side, you can
go to the casino like someone else said
check out the architecture. Buffalo is known as one of the best cities in terms of architecture. Check out some of the Frank Lloyd Wright houses. The Darwin Martin house isn't quite finished yet(they are doing a $23million renovation to it) but you can still take tours of it.
check out the Albright Knox art museum. It is one of the top art museums in the US and I have also read that the Albright-Knox possesses one of the finest collections of modern and contemporary art(I'm pretty sure admission is free on fridays)
Buffalo has a nice theater district as well
Try heading down to the Chippewa strip at night
Try skiing. Holiday Valley and Kissing Bridge south of Buffalo
If you're into hockey, go check out a Sabres game(if you can get tickets)
Shopping at the Fashion Outlets(in Niagara Falls) or the Walden Galleria
Make sure you check out the food. Don't just go to Burger King or Pizza Hut, WNY has great food. You can visit the Anchor Bar and see where "Buffalo" wings were invented(we just call them wings) WNY also has great pizza. La Novas(ranked number one independent pizza place by Pizza & Pasta Magazine) and Bocce club are my favorites. Ted's has is a great place to go for a hotdog. Also try beef on weck and a fish fry!

Sorry, that's kinda a bad time to visit. I've been there in February, and between the sleet and icy spray pelting my face, and the 'ghost-town' look of the place, there was nothing fun to do. The casinos are open, but I would rather go in the spring or fall. There's Seneca casino on the New York side and Niagara Falls Casino on the Canadian side.
Avoid summer too - high season, expensive and sticky smoggy weather.
But if you're a Southerner who has never seen snow before, then you will have a blast being chilled to the bone. (It's actually not that cold but very damp and very windy and crappy)

That's funny, I also am going to be in Buffalo for a vacation with my family in January. Hopefully, if there's snow, we will look forward to some cross country skiing. It really depends upon the kinds of things you like to do Contact the Buffalo Visitors Center for what's available in the winter months. annoying selection. The great apple area is nearly a ghetto (different than the park by making use of the falls) and the ON area is nearly a carnival. i assume the water parks at super Wolf motel and Fallsview on the Canadian area could win me over, yet fairly actually, i've got purely long gone to Niagara Falls a handful of circumstances in the 17 years i've got lived in southwestern Ontario. The falls themself are suitable to be sure, however the cities on the two area of the border are actually not. j0e
