This goes back to the bible. The concept is that poor people have a certain humility especially as it relates to material wealth. It's a recognition that all wealth is God's wealth and that we are humble servants to God. In contrast, rich people tend to be scornful, prideful, and arrogant. Here's the key point, you don't have to be poor to be poor in spirit, i.e. even rich people can have the spirit of humility and service to God that a poor person is more likely to exhibit.

I think it is a reference to humility specifically the absence of ego. Jesus came to bless the poor, rejected and down trodden. To be poor in spirit means ones proverbial cup is less than full. The idea being that the less of the world one has the more room one has to receive spiritual gifts from God.
