I want to enlarge an existing latch hole by only 2 or three millimeters in diameter. Can I use a spade bit for that?

Personally for 2 millimeters I would use a round file.

Yes if you first glue in a piece of wood neatly to hold the centre tang of the spade bit, otherwise it will only cut a ragged hole. Let the glue set overnight otherwise the wood will just spin!

I tend to use a Forstner bit as a better alternative, but these are usually more expensive, unless you buy a short one that kitchen fitters use for the hinge holes.

I would probably use a chisel to enlarge the hole. If you are talking about the hole in the door frame that the door latch goes into, then I would definitely use something like a small chisel to chip out the hole.
